Özet:
Reasons such as the increase in global seed trade and the facilitation of
international transportation ensure the rapid spread of newly emerged viruses and the
spread of diseases to clean areas. One of these viruses is Tomato brown rugose fruit virus
(ToBRFV) disease, which has gained much importance especially in recent years.
ToBRFV, It is a member of the Tobamovirus group, which also includes plant viruses
such as TMV and ToMV. While ToBRFV is transmitted by seeds over long distances, it
can spread very easily by contact between short distances. Active seed trade and the
importance of the region we are in in agriculture are the reasons that make the
transmission of ToBRFV by seed dangerous. This situation has brought the eradication
studies against the viruses found in the seeds to the fore in the fight against ToBRFV.
Within the scope of this thesis, some chemical and physical methods applied in
the disinfection of seeds were examined. Of the physical methods, the virus density was
found to be the least in seeds subjected to heat treatment at 72 °C for 72 hours. It was
found that in the application of heat treatment and UV-C applications (254 nm), the virus
loses its pathogenicity. According to the information obtained as a result of different
disinfectant applications to seeds: 1% Tsunami 100, 1% Biocon A and 10% TSP have
been determined as the optimal methods of combating this virus. In addition to the success
of seed eradication, these methods do not adversely affect germination. These studies
aimed at limiting the spread of the virus will make an important contribution to domestic
and national seed production.
