Tomato spotted wilt
virus (TSWV) in
tomato and pepper cultivation in Antalya has caused serious problems with the
breaking of resistance genes. In order to understand the cycle of this disease,
which is common in the region, two pepper and tomato greenhouses in Serik and
Elmalı districts with intense TSWV infection were observed at regular intervals
between 2019-2021. The role and importance of weeds were investigated. During
the surveys, 270 weeds and 112 different plants thought to be infected with the
virus were collected. As a result of serological studies, a wide variety of
symptoms such as mosaic, chlorotic spots, yellowing, deformity, stunting and
feeding traces of thrips were observed on the leaves of some weeds infected
with the virus, while no symptoms were observed in some infected weeds. The
highest infection rate among the infected weeds was determined in the Asteraceae family and Sonchus asper (L.) species. This family
was followed by Poaceae and Solanaceae. The strain of the virus in
infected weeds was determined as a result of qRT-PCR and Melt-curve analysis
using a special primer pair designed during the study, which could detect the
point mutation in the medium segment of the old isolate (TSWV-NRB) that did not
break resistance in tomato. As a result of the comparison of the isolates, two
different Tm values were determined. After these findings were confirmed by
sequence data, it was revealed that the kit, protocol and primers used
constitute a fast and reliable diagnostic kit for the identification of
isolates that break Sw-5 and Tsw resistance, without the need for sequencing
service.
