Abstract

Aleafcurldiseasehasbeenobservedfrequentlyingreenhouseswherecucumber(Cucumis sativus),melon(Cucumis melo)andsquash(Cucur-bita pepo) are produced in Antalya since 2019. The infected plantsshowed symptoms including apical leaf curl, vein swelling, severelystunted,wrinkledleaves,fruitshapeandtexturedisorderandacon-spicuousmosaicpattern(Figure1–5).ThesesymptomsaresimilartothosecausedbyTomato leaf curl New Delhi virus(ToLCNDV),animpor-tantmemberofthegenusBegomovirus,familyGeminiviridae.ToLCNDVistransmittedbyBemisia tabaciinapersistentcirculativemanner,caus-ingseriousdamagetoawidevarietyofplantspecies,especiallyintheSolanaceaeandCucurbitaceaefamilies(Briddon,2015).ToLCNDVwasfirstidentifiedonsolanaceouscropsinIndia(Padidametal.,1995)butthe virus is recognised as an emerging threat worldwide due to theFIGURE1TypicalTomato leaf curl New Delhi virussymptomsonsquashleavesThisisanopenaccessarticleunderthetermsoftheCreativeCommonsAttributionLicense,whichpermitsuse,distributionandreproductioninanymedium,providedtheoriginalworkisproperlycited.©2023TheAuthors.New Disease ReportspublishedbyBritishSocietyforPlantPathologyandJohnWiley&Sons.severityofthediseaseanditsincreasingprevalence.OutbreakshaverecentlyoccurredincountriesinAfrica,AsiaandEurope(EuropeanandMediterraneanPlantProtectionOrganization,2023).SampleswerecollectedfromeightadjoinedgreenhousesinAntalya,fourgrowingcucumber,twomelonandtwosquash.Threeleafsamplesandonefruitsamplewerecollectedindividuallyfromadiseasedplantineachofthegreenhouses,thatis32samplesintotal.TotalnucleicacidwasextractedfromtheleafandfruitsamplesandanalysedbyPCRwithspecificToLCNDVprimers(ToA1-FandToA1-R)whichamplifya504bpfragmentofviralDNA-A(Saezetal.,2016).Thefollowingcon-ditionswereusedforPCR:95◦Cfor5min;35cyclesof30sat95◦C,45sat55◦Cand45sat72◦C,followedbyafinal72◦Cextensionstepfor10min.SequenceswereobtainedfromsamplesfromeachoftheFIGURE2TypicalTomato leaf curl New Delhi virussymptomsonsquashfruitsNew Dis Rep.2023;47:e12180.wileyonlinelibrary.com/journal/ndr21of2https://doi.org/10.1002/ndr2.12180
