7th International Symposium on In Vitro Culture and Horticultural Breeding, Ghent, Belçika, 18 - 22 Eylül 2011, cilt.961, ss.153-157, (Tam Metin Bildiri)
Asparagus is a dioecious vegetable. The main target of asparagus breeders is to obtain hybrids composed of exclusively male plants, since they are more productive than female ones. Anther culture is a method to obtain haploids that can be doubled with colchicine treatment. In order to determine the optimal microspore development stage for androgenesis, anthers were stained with ethidium bromide and examined under fluorescence microscope. Anthers with microspores at the late uninucleate stage were cultured on Murashige and Skoog (MS) basal medium supplemented with three growth regulators at different concentrations (1 mg L-1 2.4 D + 0.1 mg L-1 BA + 0.5 mg L-1 NAA; 1 mg L-1 BA + 5 mg L-1 NAA; 0.5 mg L-1 BA + 1 mg L-1 NAA). The highest number of embryos (8.33%) and the number of plantlets (13.33%) were obtained on MS medium with 0.5 mg L-1 BA and 1 mg L-1 NAA.
