Özet

Allium junceum subs. tridentatum is an endemic bulbous plant that has the potential to be used as an ornamental plant and requires conservation in wild populations. The effects of various plant growth regulators on the regeneration capacity of immature and mature embryos were investigated in vitro. Embryos of mature and immature seeds were propagated by tissue culture; callus, shoot and bulb formation percentages (%), shoot and bulb formation per explant (number), callus weight (g) and shoot length (mm) were determined. Although mature embryos gave better results in terms of shoot and bulb formation percentage (100%, 90%, respectively, on MS media containing 1 mg/L BAP + 0.5 mg/L IAA), immature embryos formed more shoots (8.53) and bulbs (3.33) per explant than mature embryos on MS medium containing 4 mg/L BAP + 1 mg/L IAA. In this study, plants ready to be transferred to soil were obtained in quite a short time and it is thought that the method could be used to produce other commercial Allium species.
