Determination of Polymorphism in the Ovocalyxin-32 (OCX32) Gene in Different Six Brown Layer Lines Using PCR-RFLP Method

Abstract

Eggshell quality is an important trait for commercial egg industry. Strong eggshell is important to ensure that eggs can reach their final destination without damage. Ovocalyxin-32 (OCX32) is a matrix protein which is expressed at high levels in the uterine and isthmus regions of the oviduct. There are reports in literature some SNPs in OCX-32 gene associated with eggshell quality traits in chickens. The aim of the present study, was to determine polymorphism the OCX-32 gene in different six brown layer lines (Rhode Island Red – (RIRI and RIRII), Barred Rock – (BARI and BARII), Columbian Rock – (COL) and Line 54 – (L-54)) by using PCR-RFLP method. In this study, all populations were found polymorphic for OCX32 gene. While the frequency of A allele ranged from 0.346 (L-54) and 0.887 (BARI), the frequency of C allele were found between 0.113 (BARI) and 0.654 (L-54) in brown layer pure lines for OCX-32 gene. The highest genotpye frequencies for AA, AC and CC were calculated as 0.775 (BARII), 0.50 (RIRI), 0.462 (L-54) in pure lines, respectively. According to applied chi-square test, all populations were in Hardy–Weinberg equilibrium (p<0.05).
